高效液相色谱-光电二极管阵列检测器法同时测定柿蒂中乌索酸和齐墩果酸含量 被引量:6

摘要 目的采用超声辅助提取,建立高效液相色谱-光电二极管阵列检测法测定柿蒂中乌索酸和齐墩果酸的含量。方法色谱柱为Kromasil C18柱(4.6mm×250mm,5μm),流动相为甲醇:水:磷酸(体积比87:13:0.1),流速0.8ml·min^-1,检测波长210nm,柱温28℃。结果乌索酸进样量在0.359~5.744μg时,与峰面积呈良好的线性关系(r=0.999),平均回收率为98.3%,RSD为2.0%(H=6);齐墩果酸在0.174~2.784μg时,与峰面积呈良好的线性关系(r=0.999),平均回收率为99.6%,RSD为2.5%(H=6)。结论方法操作简便、快速、准确,可用于柿蒂中乌索酸和齐墩果酸含量的测定。 Objective To develop an HPLC method with photodiode array detector(PAD) for the determination of ursolic acid and oleanolic acid in Diospyros kaki by uhrasowithnic wave aided method. Methods The separation was achieved on a Kromasil C18 column (4.6 mm×250 mm, 5 μm) at 28 % using methanol - H2O - phosphoric acid (87: 13:0.1 ,V/V) as the mobile phase at a flow rate of 0.8 ml · min^-1, the detection wavelength was 210 nm. Results Ursolic acid showed good relationship at the range of 0. 359 - 5. 744 μg ( r = 0.999). The recovery was 98.3 % and RSD was 2.0 %. Oleanolic acid showed good relationship at the range of 0. 174 -2.784 μg (r = 0.999). The recovery was 99.6 % and RSD was 2.5 %. Conclusion The proposed method is simple, rapid and accurate. It can be used to determine the contents of ursolic acid and oleanolic acid in Diospyros kaki.
