Study of ^(99m)Tc-D_2C_5 in Targeted Diagnosis in Human Hepatocellular Carcinoma in Receptor Imaging

摘要 目的:探讨99mTc-D2C5用于人肝癌细胞系SMMC-7721裸鼠原位种植肝癌模型肿瘤受体成像的价值。方法:人肝细胞癌SMMC-7721采用隧道包埋法植入Balb/c nu/nu裸鼠肝左叶建立原位种植肿瘤模型。荷瘤裸鼠16只,随机分为2组,每组8只,分别经尾静脉注射99mTc-D2C5、99mTc-mIgG,放射剂量为14.8MBq。采用SPECT采集99mTc-D2C5和99mTc-mIgG注射后1h、3h显影图像。显像结束后处死动物,采用γ计数器检测体内放射性分布。结果:注射99mTc-D2C5后1 h时,裸鼠肝肿瘤部位显影,3h时肿瘤显影最清晰。注射99mTc-mIgG后,肿瘤部位未见明显放射性浓聚;3h时99mTc-D2C5裸鼠体内靶/非靶组织的比值中,肿瘤/血(T/B)为6.8,肿瘤/肝(T/L)为2.8,肿瘤/肌肉(T/M)为17.3。结论:研究表明99mTc-D2C5可与人肝癌细胞SMMC-7721高特异性和高亲和力的结合,在肝癌显像中应用前景十分乐观。 Objective: To evaluate the effect of tumor receptor imaging with ^99m Tc-D2C5 in nude mice bearing human hepatocellular carcinoma SMMC -7721 and explore the feasibility of ^99m Tc-D2C5 for using as a receptor imaging agent. Method: Human hepatocellular carcinoma cells SMMC -7721 was transplanted into the left lobe of the liver of Balb/c nu/nu nude mice to establish a hepatoma model. Sixteen nude mice bearing SMMC - 7721 were randomized into 2 groups (n = 8) to receive intravenous injection of ^99m Tc-D2C5 and ^99m Tc-mIgG via caudal vein, respectively, all at the equivalent dose of 14. 8MBq. Autoradiogram of ^99m Tc-D2C5 and ^99m Tc-mIgG in nude mice bearing SMMC -7721 xenografis were investigated lh, 3h after injection by SPECT (Single - photon emission computed tomography). All mice were sacrificed after imaging for biodistribution observation by γ counter technologies. Result: Hepatoma in nude mice were visible at lh and distincted at 3h post injection of ^99m Tc-D2C5 ,while there was no special radioactivity concentration in tumor after injection of ^99m Tc-mIgG .At 24h post injection, the ratios of tumor/blood, tumor/liver and tumor/muscle were 6.8, 2.8 and 17.3, respectively. Conclusion: The present study indicates that ^99m Tc-D2C5 has good specificity and high affinity of binding to human hepatocellular carcinoma tissues SMMC -7721. It may be used in the targeting diagnosis of liver cancer.
