金银花水提物对卵清蛋白过敏反应预防作用的研究 被引量:8

Preventable effects of the lonicera extract from water solution on intestinal microflora of the ovalbumin-induced BALB/c Mice

摘要 目的观察金银花水提物对卵清蛋白(ovalbumin,OVA)介导的速发型过敏反应有无预防作用。方法选用BALB/c雌鼠64只,建立随机分为预防组(Y组)、治疗组(T组)、激发组(Ch组)和生理盐水对照组(NS组)。Y组、T组根据灌服金银花水提物的不同浓度随机分为高(H100%)、中(M50%)、低(L25%)组,检测小鼠体内血清OVA特异性IgE;做小鼠小肠切片HE染色及甲苯胺兰染色。结果①H、M浓度金银花水提物可使YH、YM和TH、TM组致敏小鼠血清中OVA特异性IgE水平降低至正常水平;②HE染色和甲苯胺兰染色可见YH、YM和TH、TM组的小鼠小肠绒毛上皮细胞局灶性坏死、脱落,固有层淋巴细胞、浆细胞等炎症细胞浸润、小肠肥大细胞聚集和脱颗粒现象,均较Ch组显著缓解;结论金银花不能完全预防OVA介导的小鼠速发型变态反应。 [Objective] To investigate the preventable effects of the Lonicera ext on intestinal microflora of the ovalbulmin-induced BALB/c mice. [ Methods] Female BALB/c sensitized mice were randomly divided into preventive group (group Y ) , therapeutic group(group T) or the positive control group(group Ch) . The normal mice were set as the negative control (group NS). Group Y and group T were fed with 100% of the Lonicera ext as group Ya and Ta, given 50% the Lonicera ext as group YM and TM , and fed with 25% the Lonicera ext as YL and Tt. The levels of the OVA-specific IgE in serum of the mice were measured , and the tissues of small intestine were histologically examined by either HE and toludine blue stain. [Results] (1)The levels of OVA-specific IgE in serum in group Y and T were reduced after being treated by gavage with 100% or 50% of the Lonciera extract,the significant inflammatory reactions were inhibited in the mice of group YH, YM ,TH and TM. (2)The accumulated and degranulated mast cells in lamina propria were statistically reduced in the mice of group YH, YM, TH and TM by gavage with 100% or 50% of the Loncera ext. [Conclusion] The Lonicera ext OVA-induced allergic response in mice could not be prevented by the Lonicera ext.
