摘要
目的观察狼疮定颗粒对系统性红斑狼疮(SLE)患者外周血淋巴细胞(PBLC)凋亡及其[Ca2+]i的影响。方法55例女性SLE患者随机分成中西药组(28例)和西药组(27例)。西药组口服强的松为主治疗,中西药组在西药基础上加狼疮定颗粒治疗。两组连续治疗3个月,比较两组治疗前后PBLC在体外培养不同时间的凋亡率,以及[Ca2+]i水平变化。结果SLE患者PBLC的0时相凋亡率明显降低,但随体外培养时间延长而增加速率变大,SLE患者PBLC的[Ca2+]i下降;中西药组能明显改善SLE患者PBLC凋亡的异常,升高[Ca2+]i水平,且与西药组比较差异有显著性意义(P<0.05)。结论狼疮定颗粒在调整SLE淋巴细胞凋亡异常方面与西药强的松有很好的协同作用,其作用机制可能与其能促进SLE患者PBLC胞外钙内流作用有关。
Objective To observe the effect of Langchuangding Granule (Granule for Lupus Erythematosus) on the apoptosis of peripheral blood lymphocyte (PBLC) and its [Ca^2+]i of systemic lupus erythematosus (SLE) patients. Methods The 55 SLE patients were randomized into Western+Chinese Medicine group (28 cases) and Western Medicine group (27 cases). The Western Medicine group was mainly given prednisone for oral taking and the Western+Chinese Medicine group given prednisone and Herbal Langchuangding Granule for oral taking. Both groups were treated for 3 months in succession and observed on the apoptosis rate of PBLC in vitro at different period of time and the changes of [Ca^2+]i before and after treatment for comparison. Results The apoptosis rate of PBLC was obviously decreased at 0 time phase, but with the time going on in culture in vitro it became increased and level of [Ca^2+]i decreased. In Western+Chinese Medicine group, the impaired apoptosis of PBLC was obviously improved and [Ca^2+]i increased, compared with those of Western Medicine group, the difference was significant (P〈0. 05). Conclusion In the regulation of impaired lymphocyte apoptosis of SLE patients, the Langchuangding Granule has synergetic effect with prednisone, and the mechanism may be related to its promotion on inward flowing of calcium from outside of lymphocyte.
