脊髓损伤大鼠骨髓基质细胞移植后BDNFmRNA、GDNFmRNA及PLPmRNA的表达 被引量:3

Expression of BDNF,GDNF and PLPmRNA following BMSCs Transplantation in Rats with Spinal Cord Injury

摘要 目的:研究骨髓基质细胞(BMSCs)移植对大鼠脊髓损伤(SCI)后脑源性神经营养因子(BDNF)、胶质细胞源性神经营养因子(GDNF)、髓鞘前脂蛋白(PLP)基因表达的影响,探讨BMSCs促进SCI修复的机制。方法:Wistar大鼠42只,随机分为移植组(A组)和对照组(B组)各18只,制成SCI模型,将培养的BMSCs注入移植组损伤的脊髓内;另6只为假手术组(C组)。结果:术后24和72h、7d,RT-PCR半定量分析显示A、B组BDNFmRNA、GDNFmRNA和PLPmRNA表达在24及72h、7d时均明显高于C组(P<0.01),与B组比较,A组升高更显著(P<0.05)。结论:BM-SCs移植可通过促进神经营养因子的表达和轴突髓鞘化等机制促进SCI修复。 Objective; To explore the influence of bone marrow stromal cells (BMSCs) transplantation on the expression of BDNF, GDNF and PI.P mRNA in rats with spinal cord injury. Methods: Thirty six rats were divided into transplantation group and control group. Twenty-four h, 72 h and 7 days after BMSCs transplantation or operation, the expression of BDNF, GDNF and PLP mRNA was detected by using RT PCR. Results.. The expression levels of BDNF, GDNF and PLP mRNA in transplantation group were significantly higher than in control group in 24 h, 72 h and 7 days. Conclusion: The transplantation of BMSCs can elevate the expression levels of some neurotrophic factors and increase the regeneration of myelin sheath in rats with spinal cord injury.
