刺五加注射液对大鼠坐骨神经损伤的作用及可能机制 被引量:2

The effect and possible mechanism of acanthopanax senticosus injection on sciatic nerve injury of rats

摘要 目的研究刺五加注射液对大鼠坐骨神经损伤的机制及可能机制。方法 SD大鼠随机分为正常组、模型组、治疗组1(20mg/kg·d)和治疗组2(40mg/kg·d)。大鼠左侧坐骨神经夹持法建立坐骨神经损伤模型,手术当天开始腹腔注射给药,连续6周。观察大鼠左足皮肤温度、坐骨神经功能指数(SFI)和坐骨神经传导速度(SNCV)及神经组织形态学的改变,免疫组织化学染色法及Western Blot观测大鼠坐骨神经生长因子的表达。结果治疗组大鼠皮肤温度、坐骨神经功能指数、坐骨神经传导速度及神经组织形态学改变均优于模型组,神经生长因子的表达水平明显高于模型组。结论刺五加注射液对损伤后大鼠坐骨神经有促进修复的作用,其机制可能与促进神经生长因子的表达有关。 Objective To study the effect and possible mechanism of aeanthopanax senticosus injection on the sciatic nerve injury in rats. Methods Eighty SD rats were randomly averagely divided into normal group, model group, treatment group 1 (20mg/kg.d), treatment group 2(40mg/kg.d). The left sciatic nerve of rats was damaged with chucking method to establish the sciatic nerve injury model, and at same time, acanthopanax senticosus was injected peritoneally and animals were killed 6 weeks later. The temperature of left foot skin and sciatic nerve function index(SFI) and the sciatic nerve conduction velocity(SNCV) were determined. The morphological changes of nerve tissue were observed by HE staining, the expression of nerve growth factor was detected by immunohistochemical staining and Western blot, Results Aeanthopanax sentieosus injection (20mg/kg.d, 40mg/kg.d) upregulated the rat skin temperature and promoted the function recovery of sciatic nerve, accelerated the conduction velocity of sciatic nerve, increased the expression level of nerve growth factor. Conclusion The repair effect of aeanthopanax senticosus on sciatic nerve injury might be related to the upregulation of nerve growth factor expression.
