一株肾型鸡传染性支气管炎病毒的初步分离鉴定 被引量:2

Preliminary Isolation and Identification of Nephropathogenic Stram of Avian Infectious Bronchitis Virus

摘要 从哈尔滨市某肉鸡养殖场疑似传染性支气管炎的病死鸡中分离到1株肾型IBV,并对其进行鸡胚矮小化、血凝性、电镜下特征、新城疫干扰试验、致病性等生物学鉴定和N基因的RT-PCR鉴定。结果表明,该病毒分离株在鸡胚上传至第四代(F)4开始出现死亡或侏儒胚;病毒不凝集鸡红细胞;透射电镜下可见多呈球形、直径约80~120nm的病毒粒子,具有冠状病毒的典型形态特点;该病毒可干扰新城疫LaSota株在鸡胚中的增殖;将分离毒第4代尿囊液接种于6日龄雏鸡,7d后开始出现死亡,死亡率高达67%(6/9),病死鸡剖解后可见肾脏明显肿大、苍白,具有传染性支气管炎的典型病变;分离毒第5代尿囊液经N基因特异性RT-PCR获得大小约438bp的目的片断。初步确定所分离病毒为肾型IBV。 A strain of Nephropathogenic avian Infectious bronchitis Virus was isolated from died chickens that they were suspected to be infected by avian infectious bronchitis from a broiler flocks farm in Harbin city.Biological characterizations of the virus were investigated by dwarfing embryonated chicken eggs,haemagglutination test and electron microscope morphological observation,NDV interference test,virus pathogenicity to healthy chickens and RT-PCR of N gene.Results showed that the infected embryos of F4 began to die or form dwarf embryo;and that Virus could not agglutinate chicken's red blood cells;and that the virus particle was approximately 80 to 120 nm in diameter,with typical morphologyof Coronavirus;and that the replication of NDV LaSota strain was affected in chicken embryo infected by the strain;and that a 438-bp DNA fragment from the N gene was amplified by RT-PCR.The virus isolated was primarility identificated as infectious bronchitis virus.in addition,six days of chickens were inoculated with the F4,post inoculation 7 days,some of chickens began to die,the mortality rate as high as percentage 67(6/9),The kidney of the death chickens is swell and mottle prominently,with typical pathological changes of infectious bronchitis.All of these demonstated that the stain isolated from broiler fowls was nephropathogenic infectious bronchitis virus
