摘要
目的:探讨外源性神经生长因子对内源性神经生长因子的影响。方法:采用大鼠坐骨神经硅管内再生室模型,靶器官内注射神经生长因子(实验组)或生理盐水(对照组)2周。注射后1天、1周及2周,采用新生大鼠背根神经节细胞培养,测定硅管内液神经营养活性,用化学发光免疫检测法(chemilumi-nescenceimmunoassay,CLIA)测定神经生长因子含量。结果:实验组与对照组细胞存活数及神经生长因子含量,其差异有极显著性意义(t=6.49、6.42、8.82,P<0.01)。
Objective: To investigate the influence of exognous nerve growth factor (NGF) on endogenous NGF. Methods: A silicone chamber mode for sciatic nerve regeneration in adult rats had been used. The rats were given daily injections of exogenous NGF(experimental group) or normal saline solution(control group) to the target organ for 2 weeks. After 1 day、1 week and 2 weeks,we determined the neurotrophic activities by using neonatal rat dorsal root ganglia (DRG) neuron culture and measured the NGF concentrations by chemiluminescence immunoassay (CLIA) in the fluids accumulated in the silicone nerve regeneration chambers. Results:The numbers of surviving neurons and NGF concentrations of NGF treated animals are significantly different from those of controls( t=6.49、6.42、8.82,P<0.01) . Conclusions:Adminstration of NGF by target organ increased NGF neurotrophic activities and NGF content of injured nerve site.
