外源性神经生长因子(NGF)对坐骨神经损伤后脊髓前角细胞脑源性神经营养因子(BDNF)表达的影响 被引量:2

摘要 目的:探讨坐骨神经损伤后脊髓前角细胞BDNF表达变化及外源性NGF与BDNF表达的关系。方法:采用大鼠坐骨神经损伤动物模型,分为药物组、对照组,每只大鼠又分为损伤例组(L组)和健侧组(R组)两个亚组。用原位杂交、免疫级化技术检测BDNF的表达水平,现察各组在1d、7d、14d、21d及28d时的变化。用计算机图像自动处理系统进行定量分析研究。结果:药物组损伤侧脊髓前角细胞中BDNF表达水平除1d,7d组外其余均多于对照组(P<0.05);而两组健侧脊髓前角细胞中BDNF表达水平无统计学意义(P>0.05),坐骨神经损伤后损伤侧脊髓前角细胞中BDNF表达水平在最初的3周内逐渐下降,然后开始恢复,但至第4周尚未恢复到开始时的水平;而两组健侧BDNF表达水平则无明显差异(P>0.05)。结论:外源性NGF能促进坐骨神经损伤后损伤侧脊髓前角细胞中BDNF表达水平的增加;而对健侧BDNF表达水平无明显作用。 To probe into the expressive change of BDNF and the relation between exogenous nerve factor (NGF) and resume of sciatic function after injury. Methods: Rat's model which had done of sciatic damnification was adopted. The model was divivded into medication and control groups. Every rat was also divided into injury and normal side groups. By the results of immunohistochemical stain and situ hybridization, the expression of BDNF and the change of different time (1d, 7d, 21d, and 28d) were studied and analyzed its quantification by computer figure auto-disposal system. Results: Exogenous nerve growth factor improved the expression of BDNF on rat's spinal anterior horn cells, but no effect for normal side. When sciatic nerve fibers were injuried the level of BDNF in lumbar spinal motor neuron decreased gradually during the following 3 weeks and; then recovered in the 4 week, however and didn't increase normal level. Conclusion: Exogenous NGF might improve the level of BDNF in lumbar spinal motor of injury side, but it was not effective to normal side.
