外源性神经生长因子对背根神经节中TrKA及其mRNA表达的影响

摘要 目的探讨靶肌肉注射外源性神经生长因子对背根神经节中TrKA及TrNA表达的作用.方法 大鼠坐骨神经损伤后采用硅管桥接,在靶肌肉内注射外源性NGF200μ/天,术后 3、7、14及28天进行背根神经节中TrKA的免疫组织化学检查,术后7天组进行TrKA mRNA原位杂交检查.对照组采用相同的动物模型,以生理盐水进行靶肌肉注射.结果 应用NGF靶肌肉注射后,背根神经节中TrKA的表达明显增加,与对照组相比具有显著性差异(P<0.01).结论 靶肌肉注射NGF能逆转神经损伤所造成的TrKA表达下调,表明靶肌肉注射可作为NGF的有效给药途径. Aims: To investigate the effect of target muscle injectingNGF on the expression of TrKA and TrKAmRNA in rat dorsal root ganglion after the sciatic nerve injury. Method: We reparied the sciatic nerve by silicon tube after the nerve was transected and administrated exogenous NGF (200μ/day) to the target muscle. The expression of TrKA and TrKAmRNA in rat L4 L5 DRG were examined by immunohistochemistry and ISH at 3, 7, 14 and 28 days after the nerve injury. The control group was prepared by the same operation with injection of 0.9% NaCl to the muscle. Result: The expression of TrKA and TrKAmRNA incressed significantly after administration of NGF to target muscle. Significant difference were lie between experimental group and control group (P < 0.01) Conculsion: Administration of NGF to target mucle can turn off the down-regulation of the expression of TrKA and trKAmRNA after nerve injury and it is demonstrated that target muscle injecting NGF is a useful route for clinic administration.
