兔骨髓间充质干细胞与纳米晶胶原基骨修复材料体外构建组织工程骨的实验研究 被引量:8

Study of bone marrow-derived mesenchymal stem cells combined with nano-hydroxyapatite/collagen in constructing tissue-engineering bone in vitro

摘要 目的:研究兔骨髓间充质干细胞(marrow-derived mesenchymal stem cells,MSCs)与纳米晶胶原基骨修复材料(nano-hydroxyapatite/collagen,NHAC)体外复合培养的结合程度,以及构建组织工程骨的可行性。方法:分离兔MSCs体外培养、纯化,取第3代MSCs与NHAC体外复合培养,第5,10 d后扫描电镜(SEM)观察二者复合程度。结果:兔MSCs贴壁生长,增殖速度快,生长曲线符合Logistic生长曲线;兔MSCs与NHAC体外复合培养,在第5,10 d进行扫描电镜观察,发现10 d时粘附于NHAC上的MSCs细胞数(38.52±7.21)明显高于5 d时粘附的细胞数(21.28±4.70),P<0.01。结论:兔MSCs与NHAC体外复合培养结合程度较高,可以用来构建组织工程骨,为进一步的动物实验奠定基础。 Objective: To explore the feasibility of rabbit marrow-derived mesenchymal stem cells (MSCs) and nano-hydroxyapatite/collagen ( NHAC ) in constructing tissue-engineering bone. Methods: Rabbit MSCs were isolated and purified in vitro, and the rabbit MSCs of 3^rd-generation were seeded onto prepared NHAC. Their adhesion situation was analyzed by scanning electron microscope(SEM) after 5,10 days. Results: Rabbit primary MSCs and passage MSCs g-rowed adherently with great reproductive activity, and the auxodrome of serial subcultivation MSCs was in accord with Logistic auxodrome. Rabbit MSCs were combined with NHAC to construct tissue-engineering bone in vitro, and by the SEM we found that at 10th day the amount(38.52±7.21 ) of MSCs adhered with NHAC was more than it(21.28±4.70) at 5th day, P 〈 0.01. Conclusion : The adhesion degree of the compound of MSCs and NHAC was high, both of them can be used to construct the tissue-engineering bone for the secondary animal experiment.
