摘要
采用半固体和液体培养技术研究人工重组小鼠干细胞因子(SCF),观察对正常人骨髓和外周血造血祖细胞的刺激作用,发现SCF单独存在即有促增殖作用,与促红细胞生成素(Epo)联合应用有明显的协同作用。SCF可能是一种有效的刺激骨髓和/或外周血造血祖细胞增殖的造血生长因子。
The effects of recombinant mouse stem cell factor (SCF) on hematologically normal adult bone mar-row (n=21) and peripheral progenitor cells (n=11) were studied by means of semisolidified and liquified culturue technies in vitro. In semisolid culture, SCF alone could result in colony formation, moreover, SCF plus Epo could significantly increase both the number and size of colonies of CFU-GM, BFU-E and CFU-E. After the bone mar-row cells were incubated with SCF plus Epo for seven days in liguid culture, the total cell count was increased for 12. 69 held, CFU-GM 25. 07 held and CD34+cells were increased markedly. 1n addition, SCF plus Epo could stimu-late the peripheral blood progenitor cells as well. The results indicated that SCF and Epo have collaborative syner-getic effects on colony formation and CFC growth.
