Effects of 6-BA and NAA on in vitro Regeneration of Leaf Piece in Chrysanthemum morifolium

摘要 以叶片为外植体进行了菊花(Chrysanthemum morifllium)组织培养技术的研究。结果表明,菊花叶片用0.1%HgCl2消毒7min为佳;较低浓度的6-苄氨基腺嘌呤(6-BA)和较高浓度的α-奈乙酸(NAA)有利于愈伤组织的诱导,其中以MS+1.0mg/L6-BA+0.3mg/LNAA诱导的效果较好;较高浓度的6-BA和较低浓度的NAA则有利于芽的诱导,其中以MS+4.0mg/L6-BA+0.2mg/LNAA诱导的效果较好;以1/2MS附加0.1mg/LIBA的培养基诱导的根量最大,且根粗长。 Tissue culture of Chrysanthemum morifolium was studied using leaf piece as explants.The results showed that the effect of disinfection for 7 min with 0.1% HgCl2 was the best;Lower concentration of 6-BA and higher concentration of NAA benefited the induction of calli,and higher concentration of 6-BA and lower concentration of NAA benefited the induction of shoots.Among them,MS+1.0 mg/L 6-BA +0.3 mg/L NAA was optimal to calli induction and MS+4.0 mg/L 6-BA +0.2 mg/L NAA was optimal to shoots induction.1/2 MS+0.1 mg/L IBA was optimal to roots induction.
