摘要
研究表明,茄链格孢(Alternariasolani)培养滤液中存有抗菌活性物质。采用改进Bartels-keith方法对该物质进行了提取分离并进行抗菌活性跟踪。对纯品通过熔点测定、元素分析及1H-NMR、13C-NMR、1H,1H-COSY及DEPT光谱解析,证明该物质为交链孢酸(Alternaricacid)。离体试验结果表明,0.1μg/ml交链孢酸对Botrytiscinerea,Sclerotiniasclerotiorum菌丝生长抑制率分别为64.3%和73.6%。M.I.C值≤100μg/ml。500μg/ml交链孢酸对B.cinerea分生孢子萌发没有影响,但可明显抑制孢子萌发后芽管的伸长,≥10μg/ml时可完全抑制S.sclerotiorum菌核萌发。活体试验表明,500μg/ml交链孢酸对黄瓜灰霉病和油菜菌核防治效果分别为82.7%和83.3%。
It was found that there were some antifungal active substances from culture filtrate of Alternaria solani. Improved Bertels Keith method was adopted for the extraction and isolation of the substances, and the monitoring tests of antifungal activity were carried out in the separation process. It has been proved that alternaric acid was an active component by means of spectroscopic analyses of 1 HNMR, 13 C NMR, 1 H, 1 H COSY and DEPT, and by element analysis and measurement of melting point. The inhibitory rate of 0.1 μg/ml alternaric acid against hyphae growth of Botrytis cinerea and Sclerotinia sclerotiorum in vitro, were 64.3 and 73.6%, respectively. M.I.C.≤100 μg/ml. At 500 μg/ml, alternaric acid did not inhibit the germination of spore of B. cinerea , but clearly retarded the extension of the germ tubes after germination. When the concentration was ≥10 μg/ml, the sclerotia germination of S. sclerotiorum was inhibited completely. The control effects of alternaric acid at 500 μg/ml against the grey mold of cucumber and sclerotinia rot of rape on living plants reached 82.7% and 83.3%, respectively.
