摘要
本研究以耐渍型亚麻品种Y4F082和非耐渍型品种Agatha及其杂交F2代为材料,采用分离群体分组分析(Bulked Segregate Analysis,BSA)法,对亚麻耐渍基因进行RAPD分析。在500个随机引物中筛选出一个引物S1377在两亲本及抗、感基因池中均能扩增出一条大小为800bp稳定的差异条带,命名为S1377-800,通过F2代个体验证,耐渍型个体均能扩增出此差异条带而非耐渍型个体中不能扩增出此差异条带,证明S1377-800是与亚麻耐渍基因紧密连锁的RAPD标记。
In this experiment, the resistant parent Y4F082( ♂ ), sensitive parent Agatha( ♀ ) and their F2 generation were used for identification of the molecular markers linked to waterlogging tolerance gene of flax. By RAPD-BSA, 500 random primers were used to amplify the parents gene and two gene pools. When the primer was S1377, a polymorphic fragment of 800bp was found in parents and two pools, and named S1377-800. The fragment existed in the resistant plants and F2 population, it was indicated that the RAPD marker S1377-800 was closely linked to waterlogging tolerance gene in flax.
