枸杞多糖对细胞膜流动性及蛋白激酶C的体外效应 被引量:6

EFFECTS OF LYCIUM BARBARUM POLYSACCHARIDE ON THE CELL MEMBRANE FLUIDITY AND PROTEIN KINASE C IN VITRO

摘要 目的:研究枸杞多糖(LBP)体外免疫调节效应的作用机制。方法:以DPH作为荧光深剂,采用荧光偏振法观察LBP对免红细胞膜流动性的影响。并以32P-组蛋白内参入的放射性强度检测法,观察LBP对淋巴细胞胞膜和胞浆蛋白激酶C(PKC)活性的影响。结果:100mg/LLBP可明显促进兔红细胞细胞膜的流动性,并能增强10mg/L和25mg/LConA对膜流动性的促进作用。同时100ms/LLBP尚可增加ConA活化后的小鼠脾淋但细胞膜上的PKC的活性,但对胞浆PKC的活性无影响。结论:LBP的体外作用途径可能是作用于细胞膜,通过促进细胞膜的流动性及促进ConA活化的小鼠脾淋巴细胞胞浆内PKC从胞浆到胞膜的激活移位而发挥免疫调节效应的。 Objective: The immunoregulatory mechanism of LBP was investigated in vitro. Methods:The effect of Lycium barbarum polysaccharide (LBP) on the cell fluidity in rabbit red blood cell wasstudied by the fluorescence polarization technique wlth DPH fluorescence probe. Then the effect of LBPon the protein kinase C (PKC) in murine lymphocytes membrane and cytoplasm was observed by radioactivation analysis with 32P-histone incorporation. Results: 100 mg/L LBP significantly enhancedcell nuiditY (P<0. 001), and also accelerated the action of ConA(10 mg/L, 25 mg/L) on cell fluidity(P<0. 05) in the rabbit red blood cell. Alternately, 100 mg/L LBP could enhance the membrane PKCactivity (P<0. 01 ),and has no effect on cytop1asm PKC activity in lymphocytes activated by ConA.Couclusion: Action on the cell membrane and enhancement of LBP on cell membrane fluidity and PKCactivated conversion from lymphocyte cytoplasm to membrane may be invo1ved in the way of LBP toperform its immunoregulatory effect.
