摘要
目的探讨硫普罗宁(MPO)对肝豆状核变性(HLD)大鼠的治疗作用。方法用铜负荷饮食法制作HLD动物模型,将模型大鼠分为治疗组与非治疗组,治疗组给予MPO100mg·kg-1·d-1灌胃,非治疗组继续给予铜负荷饮食,对照组大鼠传统饮食喂饲。观察各组大鼠生长发育、丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、总蛋白(TP)、白蛋白(Alb)、肝铜及24h尿铜水平变化情况。结果非治疗组及治疗组ALT、AST、肝铜、24h尿铜水平均较对照组显著增高(P<0.05),TP和Alb与对照组均无显著差异(P>0.05);治疗组AST、肝铜较非治疗组显著降低,Alb、24h尿铜较非治疗组显著升高(P<0.05)。结论硫普罗宁能降低HLD模型大鼠肝组织铜水平,增加24h尿铜排泄,并能够改善肝功能。
Objective To observe the therapeutic effect of tiopronin oh hepatolenticular degeneration (HLD) in rats. Methods The rat models of HLD were set up by copper overloaded diet. The rats were divided into treatment group and non-treatment group. The rats in treatment group were giving tiopronin once a day at a dosage of 100 mg/kg and those in non-treatment group were fed with copper overloaded diet, however, the rats in control group were fed with normal diet. The state of growth,blood ALT,AST, total protein (TP) ,albumin (Alb) ,liver copper and 24 hours urine copper of these rats were observed and compared with those in control group. Results The levels of ALT, AST, copper in liver and urine copper within 24 hours in treatment group and non-treatment group were significantly higher than those in control group ( P 〈 0.05),there was no significant difference in the levels of TP and Alb between the two model groups and control group; the levels of AST and copper in liver in treatment group were obviously lower than those in non-treatment group; the levels of Alb and urine copper within 24 hours in treatment group were obviously higher than those in non-treatment group ( P 〈 0.05). Conclusion Tiopronin couht reduce the levels of copper in liver of HLD rats, and increase the evacuation of urine copper in 24 hours, and improve hepatic function.
