肝细胞微囊化移植对大鼠肝豆状核变性模型铜代谢的影响

摘要 目的观察微囊化肝细胞腹腔移植对大鼠肝豆状核变性(HLD)模型血清铜及肝铜代谢的影响。方法采用铜负荷饮食法制作大鼠HLD模型。气流法制备含肝细胞微囊,并采用腹腔移植的方法进行裸细胞或微囊化细胞移植,分别设空白对照组(Ⅰ组)、氯化钠溶液组(Ⅱ组)、裸肝细胞腹腔移植组(Ⅲ组)和微囊化肝细胞移植组(Ⅳ组),测定5个时间点大鼠HLD模型ALT、AST、白蛋白水平,血清铜、肝铜含量。结果Ⅱ、Ⅲ、Ⅳ组大鼠各时间点ALT、AST、血清铜、肝铜值均较Ⅰ组大鼠明显升高(均P〈0.01),合成白蛋白水平明显下降(均P〈0.01)。7-14天后IV组大鼠ALT、AST、血清铜、肝铜水平较Ⅲ组明显下降(均P〈0.01),最低值分别达(85.1±7.0)U/L(、87.9±13.7)U/L、(2.44±0.18)μg/mL(、26.73±3.22)μg/g,而白蛋白则于14天后高于Ⅲ组,最高达(38.36±1.52)g/L。结论肝细胞微囊化腹腔移植可明显减轻大鼠HLD的肝铜沉积,加速血清铜的代谢,可成为一种细胞移植治疗HLD的新方法 。
