摘要
应用聚丙烯酰胺凝胶垂直板电泳(PAGE)分离和研究成体泥鳅心肌、骨骼肌、肝脏、卵巢、脑、眼、血清七种组织器官中的LDH同工酶。结果表明:在电泳图谱负极端的相同部位,骨骼肌出现5条,肝脏有4条,脑组织1条LDH同工酶区带;其余多数组织中均为3条酶区带;此外,近负极端,肝脏组织还有1条其他组织所没有的LDH同工酶区带。这提示了泥鳅的LDH同工酶可能是由LdhA、LdhB和LdhC3个基因座位编码的A、B、C亚基组合成的四聚体。定量分析表明LDH同工酶活力具有明显的组织特异性:血清、卵巢、肝脏等组织中LDH5活性最强。
The electrophoretic patterns of LDH isoenzymes in serum, eye, brain,ovary,liver, skeletal muscle and cardiac muscle from adult M. anguillicaudatus distributed over bog were studied by means of polyacrylamide gel vertical electropheresis. The results indicated that in the same area near negative pole there were three bands in the most of these tissues, but one band in brain, four bands in liver and five bands in skeletal muscle respectively.In addition, in the different area near positive pole there was one band in liver. All these suggest that the LDH isoenzymes of M. anguillicaudatus might be tetraner made up by A, B and C subunit codied by three gene loci,namely Ldh-A, Ldh-B and Ldh-C. The quantitative analysis exhibited a tissue specific expression: the strongest activity of the LDH isoenzyme were LDH 5 in serum, ovary and liver, LDH 2 in cardiac muscle and LDH 3 in skeletal muscles.
