抗氧化剂对梨基因组DNA提取的影响

摘要 以鸭梨和王子廿世纪梨的幼叶为试材,采用CTAB法提取基因组DNA,分析抗氧化剂种类和浓度对DNA提取效果的影响。结果表明,用β-巯基乙醇提取DNA能有效地抑制酚的氧化,所提的DNA呈白色,OD260/OD280为1.948~2.023,平均为1.990,纯度较高,DNA产率为52.8~175.8,电泳图谱主带清晰,能满足对DNA质量要求较高的试验,其提取效果优于VC和PVP两种抗氧化剂。从抗氧化剂浓度上看,2.0%的β-巯基乙醇效果优于1.5%和2.5%的β-巯基乙醇,OD260/OD280平均为1.994,产率平均为128.3μg.g-1。 Effects of oxidation inhibitor types and there coneentrition on the the DNA extraction were studied by using the young leaf of Yali-pear and prince twenty century pear as the material and CTAB as the extracts. The result indicated that the extracting efficiency of β-mercaptoethanol (BME) surpassed VC and PVP; from the point of oxidation inhibitor concentrition, the extracting efficiency of 2.0% β-mercaptoethanol (BME) surpassed 1.5% and 2.5% β-mercaptoethanol (BME) ; the average of OD260/OD290 was 1.994, and the average production rate was 128.3 μg.g^-1.
