摘要
通过对一株耐硒酵母进行UV和60Co逐级诱变,获得一株产量高的突变株Y7,发酵液的硒总含量达19mg/L,是出发菌株的1.58倍,经多次传代试验,证明其稳定性良好。通过单因素、L16(37)正交试验对其发酵培养基和培养条件进行优化,试验表明富硒酵母发酵培养基的最佳配方是:蔗糖6%,牛肉膏1%,蛋白胨1%,K2HPO4 0.15%,亚硒酸钠35μg/L;最佳培养条件:初始pH为4.0,温度32℃,装液量60mL/250mL摇瓶,转速200r/min,10%(v/v)接种量,培养48h。富硒酵母的硒总含量达23mg/L以上,是出发菌株的1.92倍。
The selenium-enriched yeast was treated with UV, 60^Co, and the mutant Y23 was obtained. The mutant was stable. Fermentation conditions were investigated. The optimized fermentation medium determined by single factors and L16(3^7)orthogonal test was(v/v): sucrose 6%, beef extract 1%, peptone 1%, K2HPO4 1.5%. The corresponding fermentation conditions were: initial pH 4, culture temperature 32℃, 60mL liquid medium in a flask of 250mL (rotation speed of 180r/min) and inoculum concentration of 10%(v/v). Under the optimized conditions, the selenium content of the cells was up to 23mg/L.
