枸杞多糖对地塞米松诱导的小鼠脾脏细胞凋亡的影响 被引量:5

摘要 目的探讨枸杞多糖(LBP)对地塞米松(DEX)诱导的小鼠脾脏细胞凋亡的影响。方法以水提取-乙醇沉淀法制备LBP,分别给予实验小鼠每天口服LBP 25、50、100 mg,10 d后体外实验取小鼠脾脏细胞与10-5、10-6、10-7mol/L的DEX共同孵育,6 h后检测细胞凋亡率;体内实验给予小鼠腹腔注射DEX(25 mg/kg),10 h后取脾脏细胞直接检测凋亡率。结果实验动物预先口服LPB 10 d后,可使DEX诱导的小鼠脾脏细胞凋亡率显著降低,并呈现量效关系。结论LPB的免疫调节和促进作用与其对免疫细胞的保护、降低免疫细胞凋亡有关。 Objective To study the effect of polysaccharide from lycium barbarum (LBP) on spleen cell apoptosis induced with dexamethasone in mice. Methods The Kunming mice were orally administrated with 25, 50, and 100 mg LPB everyday respectively for 10 days. Then the mice were divided into two groups (A groups for in vivo experiment and B groups for in vitro experiment). Spleen cells which was separated from mice in group A was incubated with dexamethasone at the doses of 10^-5, 10^-6, and 10^-7mol/L for 6 hours, then the rates of apoptosis were detected. 10 hours after intrapefitoneal injection of dexamethasone at the dose of 25 mg/kg in mice in group B, spleen cells were separated and the rates of apoptosis were detected immediately. Results Oral administration with LBP for 10 days could significantly lower the apoptosis rates of spleen cells of mice induced with dexamethasone with dose-dependent effects. Conclusion The up-regnlation of LBP on immuno-function is related to its effects of low cell apoptosis.
