摘要
褐藻酸降解菌埃氏交替单胞菌 (Alteromonas espejiana)菌株 Al0 2发酵培养时褐藻酸酶形成条件研究表明 ,其产酶的培养基最适褐藻酸钠含量为0 .3~ 0 .6 % ,氮源为 0 .5 %的蛋白胨 ,p H7.5 ,装量是在 5 0 0 ml三角瓶中装培养基2 0 0 ml。培养温度为 2 5℃ ,培养时间为 1 44 h。利用该菌株进行发酵培养 ,制备褐藻酸酶。制备出的褐藻酸酶作用于海带和裙带菜 ,可使海带和裙带菜细胞游离产生大量的单细胞和原生质体。
The strain Al02 of Alteromonas espejiana is alginate degrading bacteria. The conditons for alginase porduction were examined. The optimal conditions for alginase production are as follows: medium consisting of 0.3 0.6% sodium alginate as carbon source and 0.5% peptone as nitrogen source, pH7.5, with 200ml fermentation medium in 500ml flask, 25℃, incubation 114h. Alginase was prepared by fermenting strain Al02. A large number of sepaerated cells and protoplasts were obtained when alginase acted on Undaria pinnatifida and Laminaria japonica.
