失血合并闭合性骨折致小鼠脾脏树突状细胞抗原递呈功能下降 被引量:7

摘要 目的研究失血合并闭合性骨折对小鼠脾脏树突状细胞(DC)抗原递呈功能的影响并初步探讨其机制。方法致伤后24小时分离小鼠脾脏DC,检测脾脏DC在体外抗原作用下刺激脾非黏附细胞增殖的能力、细胞因子分泌功能及其表面重要分子表达的情况。结果创伤小鼠脾脏DC体外刺激脾非黏附细胞增殖的能力下降,细胞因子分泌水平发生了变化,流式细胞分析结果显示其成熟度下降。结论失血合并闭合性骨折可致小鼠脾脏DC的抗原递呈功能下降。 Objective To study the antigen-presenting function of splenic dendritic cells ( DC ) in mice after hemorrhage plus closed fracture. Methods Splenic DCs were isolated 24h after hemorrhage plus closed fracture, then the ability to stimulate nonadherent splenocytes proliferation in the presence of conalbumin in vitro was detected,IL-12 and IL-10 levels in cell supernatants were measured by ELISA kits. MHC class Ⅱ , CD40, CD80, and CD86 on DC were measured using flow cytometry. Results The ability of DC to stimulate nonadherent splenocytes proliferation was decreased after injury, and the levels of cytokines were changed. The flow cytometric analysis demonstrated that the degree of maturity of DC decreased after injury. Conclusion Antigen-presenting capacity of splenic DC in mice decreased after hemorrhage plus closed fracture.
