摘要
[目的]通过对气虚血瘀大鼠模型表征及血液流变学的动态观察,探索模型大鼠敏感表征及其血液流变学特征,为气虚血瘀证生物学基础研究提供思路和依据。[方法]力竭游泳法制作气虚血瘀大鼠模型,根据拟定的表征体系进行动态(1周、2周、4周)观察,同时检测相关血液流变学指标变化。[结果]模型组大鼠在力竭游泳后(2周、4周)分别表现出体质量下降、游泳时间缩短,旷场分析中(2周、4周)中央格时间延长,舌质(1周、2周、4周)由淡红转为紫黯;血液流变学指标中,(2周)血浆黏度增高,凝血酶原时间(PT)缩短(1周、2周、4周),(4周)活化部分凝血活酶时间(APTT)缩短,(4周)纤维蛋白原含量(FIB)降低。[结论]力竭游泳所致气虚血瘀大鼠模型成立;相关表征在一定程度上能够反映气虚血瘀状态,其中精神状态差、体质量下降、游泳时间缩短、懒动等体现气虚,舌质转黯、紫黯等体现血瘀;与表征相关的血液流变学指标改变符合血瘀证一般特征,同时具有一定特征性。
[Objective] To establish a model of Qi deficiency and blood stasis model in rats and discover the sensitive external signs and the hemorheologic property. [Methods] Qi deficiency and blood stasis was induced by exhausting swim every day for a certain period (1, 2 or 4 w). External signs and hemorheology were observed. [Results] Compared with normal rats, average body weight of model rats (2 and 4 w) was less, time of swimming (2 and 4 w) was shorter, while time of staying in the middle square in open-field analyses (2 and 4 w) was longer. In model rats (1, 2 and 4 w) the tongue color turned from light red to dark or dark purple. Blood viscosity increased (2 w), PT (1, 2 and 4 w), APTT (4 w) and FIB (4 w) decreased in model. [Conclusions] Qi deficiency and blood stasis is successfully induced in rats based on the evidence of relative external signs and hemorheological property as above.
