摘要
为深入研究MT1-MMP在血管生物学中的功能,该研究比较了MT1-MMP及与其功能相关MMP-2、TIMP-2在人微血管内皮细胞株HMEC-1和人大血管内皮细胞株EAhy926、ECV304中的mRNA表达差异。实时定量PCR分析显示3株细胞均表达MT1-MMP与TIMP-2,其中MT1-MMP在EAhy926中表达最高,TIMP-2在3株细胞中呈现相似的表达量,在ECV304中相对表达较高,而仅在EAhy926中检测到MMP-2的高表达。MT1-MMP和MMP-2在典型的大血管内皮细胞株EAhy926中的高表达可能与该类内皮细胞独特的来源、表型特点和功能有关。
In order to investigate the role of MT1-MMP in vascular biology, the mRNA expression of MT1- MMP/TIMP-2/MMP-2 in human micro-vascular cell line HMEC-1,human macro-vascular cell lines ECV304 and EAhy926 were characterized.Real-time PCR analysis showed MT1-MMP and TIMP-2 were extensively expressed in all cells,among which, the highest expression of MT1-MMP was observed in EAhy926 and TIMP-2 was similarly expressed with relatively high expression in ECV304.The high expression ofMMP-2 mRNA was only detected in EAhy926.The remarkably high expression of MT 1-MMP and MMP-2 in typical human macro-vascular cell line EAhy926 may be related to its distinct origin, phenotypical characteri.stics and functions.
