摘要
目的:利用脉冲场凝胶电泳(PFGE)技术探讨O139群霍乱弧菌的分子分型方法。方法:用SfiⅠ酶和NotⅠ酶切O139群霍乱弧菌染色体DNA,经脉冲场凝胶电泳进行片段分离。结果:14株O139群霍乱弧菌经SfiⅠ酶切后电泳分离可得一个PFGE型,用NotⅠ酶切电泳可分为2个PFGE型。结论:脉冲场凝胶电泳技术分析O139群霍乱弧菌,分型发现微小差别有助于分析追踪疫情和来源。
Objectlve:To study the gene typing methods of Vibrio cholerae O139 by pulsed -field gel electrophoresis(PFGE), Methods:The chromosome DNA of Vibrio cholerae O139 was digested by Sfi Ⅰ enzyme and Not Ⅰ enzyme and the PFGE was carried out. Results: There was 1 genotype by Sfi Ⅰ enzyme and 2 genotypes by Not Ⅰ enzyme in 14 strains of Vibrio cholerae O139. Conclusion:The fine typing ability and reliability of PFGE are helpful for analyzing the epidemic situation and finding out the infectious resource of the outbreak.
