荧光定量PCR技术在小儿尿液巨细胞病毒检测中的应用 被引量:6

摘要 目的探讨荧光定量聚合酶链式反应(FQ-PCR)方法检测尿液巨细胞病毒在儿童CMV感染诊断中的意义。方法分别应用FQ-PCR和ELISA法检测75例确诊或高度怀疑为CMV感染和25例对照儿童尿CMV-DNA和血清CMV-IgM,比较病毒DNA与血清抗体的平行性和敏感性。结果75例确诊或疑诊CMV活动性感染患儿中,检测到CMV-DNA 66例;检测到CMV-IgM 54例。两种方法阳性符合率为77%,FQ-PCR和ELISA法阳性检出率分别为88%和72%,有统计学差异(P<0.05)。结论FQ-PCR法检测尿CMV-DNA对诊断活动性CMV感染具有良好的应用价值。 Objective To explore the significance of detemaining of urine cytomegalovirus (CMV) DNA in 20 -. + - 0 - diagnosis of eytomegalovirus infection in children by fluorescent quantitative polymerase chain reaction ( FQ - PCR) assay. Methods CMV DNA and anti - CMV IgM in the urine of 75 suspected CMV infected children and 25 controls were determined by FQ - PCR and ELISA. Results Out of the 75 children suspected with active CMV infection CMV DNA was detected from 66 children and CMV - IgM was detected from 54 children with the positive rates of 88% and 72%, respectively, showing significant difference ( P 〈 0.05). The positive agreement of the two methods was 77%. Conclusion FQ - PCR can be widely used in detection of CMV DNA for diagnosis of active CMV infection in children.
