摘要
目的:通过对脱蛋白骨制备过程中骨块大小、保存方法的改进及理化性能研究,探索为长骨大段缺损修复提供合适的支架材料。方法:将猪股骨下端松质骨沿骨小梁方向制成30mm×3mm×3mm大小骨块。参照改良法制备脱蛋白骨,进行深低温(-85℃)冷冻3个月,蒸馏水浸泡48 h后置入干燥箱内烘干,真空抽气、包装,60Co照射消毒,-4℃保存备用。对脱蛋白骨进行形态结构、组成成分、力学性质及细胞相容性检测。结果:本法制备的脱蛋白骨具有天然网状结构,其无机成分为羟基磷灰石,有机成分为Ⅰ型胶原,力学性能良好,有良好的细胞相容性,细胞上架率高,无明显的免疫排斥反应。结论:本法制备的猪脱蛋白骨各项性能良好,可用作长骨大段缺损修复的支架材料。
Objective:By studying the properties of deproteinized bone after preparation, we are to explore suitable scaffold for big segmental long bone defect. Methods : Bone blocks as 30 mm × 3 mm × 3 mm were prepared from inferior extremity of pig femur along bone trabecula, and processed referring to an improved method. Their morphological features, constitute components, cell compatibility, mechanical properties and immunology peculiarity were examined. Results:It showed that the deproteinized bone maintained natural network pore system. The main organic material is collagen Ⅰ and inorganic is hydroxyapatite. It has good mechanical properties, cell adhesion rate and histocompatibility. Conclusion : This deproteinized bone has many good properties and may be applicable as scaffold for long bone big segmental defect reparation.
