摘要
本文介绍用~3H-TdR掺入示踪技术,探讨白细胞介素2(IL-2)对辐射引起人血淋巴细胞增殖抑制的逆转作用。用^(60)Coγ射线体外照射人血淋巴细胞后,加入植物血球凝集素(PHA)和外源性IL-2常规培养,以淋巴细胞DNA中~3H-TdR掺入量的变化来衡量T淋巴细胞的增殖能力。结果表明,1-40Gy剂量的γ射线照射均可引起T淋巴细胞增殖力抑制,40Gy剂量组增殖率只有27.6%;加IL-2后,1-10Gy剂量组的T淋巴细胞增殖幸均明显增加,提示IL-2有使受辐射损伤的T淋巴细胞增殖能力恢复的效应。
The present study is to ascertain the offect of IL-2 onrecovery of proliferation ability in inradiaced r lympnocytes.~3H-thymidine(~3H-IdR)incorporation assay was used to evaluated the proliferationability in human petipheral blood T Lymphocytes,irradiated by variousdoses of Co gainina rays with or without exogenous IL-2 added.Afterexposure to gamina rays of 1,2.5. 5, 10, 2v and 40 Gy,the DNAsynthesis was found to bo blocked.It indicated tne manked damage of theproliferation ability of T cells.The rates of ~3H-thymicine incorporationinto DNA of cells decreased with increasing doses.~3H-TdK incorporationin lymphocytes IrradiatEd at dosE ranged from 1 to 10Gy,was lessthan that in non-irradiatEd ones.The lowest of proliferation.rates was27.6%.This inhibition of proliferaton was parcially restored by adding IL-2,but the inhibitory effect (exposure to doses of more than 10 Gy) on theproliferation of human peripheral blood lymphocytes was not reversed.
