mRNA expression of GLI1 in pancreatic carcinoma and clinical significance thereof

摘要 目的探讨胰腺癌组织中 Hedgehog 信号通路成员 GLI1 mRNA 的表达情况及其临床意义。方法采用逆转录聚合酶链反应(RT-PCR)检测25例胰腺癌组织及其相应近癌旁组织、癌旁正常组织 GLI1 mRNA 的表达,并分析胰腺癌组织中 GLI1 mRNA 表达与肿瘤大小、分化程度及转移的关系。结果 GLI1 mRNA 在胰腺癌组织、近癌旁组织及癌旁正常组织中的阳性表达率分别为68.0%(17/25)、24.0%(6/25)、0;胰腺癌组织 GLI1 mRNA 阳性表达率显著高于近癌旁组织及癌旁正常组织(均 P<0.01);GLI1 mRNA 阳性表达率与肿瘤分化程度相关(P=0.014),与肿瘤大小、有无淋巴结及远处转移无关(P>0.05)。结论 GLI1 mRNA 在胰腺癌组织中有较高的阳性表达率,且其表达率与肿瘤分化程度相关。检测 GLI1表达有助于胰腺癌的诊断,可能作为判断胰腺癌恶性程度及预后的指标。 Objective To investigate the mRNA expression of GLI1, a transcription regulator of Hedgehog signaling pathway, in human pancreatic carcinoma and to explore its clinical significance. Methods Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the mRNA expression of GLI1 in the tumor tissues, tissues near tumor, and normal tissues obtained during operation from 25 pancreatic carcinoma patients. Results The GLI1 mRNA expression rate of the tumor tissues was 68.0% (17/25), significantly higher than those of the tissues near tumor,and normal tissues [24.0% (6/25) and 0 (0/25) respectively, both P 〈0.01 ]. The GLI1 mRNA expression rate was significantly associated with the differentiation degree of tumor tissue (P = 0. 014 ), and not significantly associated with the tumor size, invasion, and metastasis ( all P 〉 0.05 ). Conclusion GLI1 mRNA expression is strong in pancreatic carcinoma tissues and is significantly associated with the differentiation degree of tumor tissue. With diagnostic implication, GLI1 mRNA expression may be regarded as a parameter of determining the degree of malignancy and prognosis of pancreatic carcinoma.
