结直肠癌肿瘤标记物的筛选及其表达变化 被引量:6

Screening and expression of tumor markers in colorectal carcinoma

摘要 目的筛选结直肠癌肿瘤标记物,探讨其在术前、术后患者血清中的表达变化。方法用表面增强激光解吸电离飞行时间质谱(SELDI-TOF-MS)技术检测87例结直肠癌、68例结直肠良性病、56例正常人血清中的差异蛋白,进而检测87例结直肠癌术前及术后15 d 内血清中的差异蛋白,从而筛选出结直肠癌肿瘤标记物。结果筛选出质荷比为5955Da、5972Da 的2种蛋白,与结直肠良性病组和正常人组比较,其在结直肠癌患者血清中高表达(P<0.01);与结直肠癌术前组比较,术后1 d其表达水平明显增高(P<0.01),术后4 d 已经降至术前水平(P>0.05);与术后1 d 组比较,术后4 d 至15 d 其表达水平基本处于下降趋势(P<0.01),但术后15 d 仍未降至正常水平(P<0.01)。结论质荷比为5955Da、5972 Da 的2种蛋白可作为结直肠癌肿瘤标记物,其在术前、术后15 d 内血清中的表达有一定的变化规律。 Objective To screen the tumor markers of colorectal carcinoma and to investigate their expression in preoperative and postoperative serum. Methods The distinct proteins in serum were detected in 87 cases of colorectal carcinoma, 68 cases of benign colorectal diseases and 56 healthy people by surfaceenhanced laser dosorption/ionization time-of-flight mass spectrometry (SELDI-TOF-MS). The distinct proteins in serum were detected in 87 cases of colorectal carcinoma pre-operation and within 15 days post operation with the same methods. Results Two proteins with mass-to-charge ratios of 5955Da and 5972Da were screened. Compared with benign colorectal diseases and healthy control, the expression of the two proteins was obviously up-regulated in colorectal carcinoma( P 〈 0. 01 ). Compared with pre-operation, the expression on the 14 day post operation was obviously up-regulated ( P 〈 0. 01 ), and the expression decreased to pre-operative level on the 4^th day post operation. The expression of the two proteins turned out to be a descendent tendency form the 4^th to 15^th day post operation, but did not reach the normal level as found in healthy control. Conclusions Two proteins with mass-to-charge ratios of 5955Da and 5972Da could be regarded as tumor markers in colorectal carcinoma, the expression may has some regularity.
