摘要
目的:应用表面增强激光解吸电离飞行时间质谱(SELDI-TOF-MS)技术寻找乳腺癌患者血清特异性蛋白。方法:应用弱阳离子蛋白质芯片(WCX2)及SELDI-TOF-MS技术检测12例正常人、10例乳腺良性病患者和22例乳腺癌患者血清中蛋白的相对含量。结果:三组样本质荷比在2000~30000Da间3939.314Da与3960.478Da的2个蛋白峰有显著差异。结论:SELDI-TOF-MS技术对乳腺癌早期诊断和特异性肿瘤标记物的筛选等方面具有一定价值,值得推广。
Objective: To detect the serum proteomic patterns in breast cancer patients by surface enhanced laser desorption/ionization time of flight mass spectrometry (SELDI-TOF-MS). Methods: The relative contents of serum proteins of 12 healthy people,10 patients with benign breast diseases and 22 patients with breast cancer were detected by WCX2 chip and SELDI-TOF-MS. Results: At the M/Z values range from 2000Da to 30000Da, the contents of two proteins (3939.314Da and 3960.478Da)had significant difference in the three groups. Conclusions: SELDI-TOF-MS shows great potentiality for breast cancer in early diagnosis and screening of tumor biomarkers, and deserves generalization.
