摘要 本文讨论了一种灵敏、高选择性的方法分别测定葡萄糖和胆固醇,并介绍了成二醛固定化酶(葡萄糖氧化酶或胆固醇氧化酶)后产生的过氧化氢借助于鲁米诺-铁氰化钾发光反应由流动注射法测定。对血清中样品的前处理和反应试剂浓度的影响也分别作了研究。本法有良好的线性工作范围,连续进样100次仍能保持工作的稳定性,相对标准误差为3.8~5.2%。 A sensitive and highly seletive method for the determination of glucose and cholesterol was described individully, and a immobilization method using glutaric dialdehyde was introduc- ed. Glucose or cholesterol passed through a immobilized enzyme (grucose oxidase or cholesterol oxidase), the generated hydrogen peroxide was detected with luminol-potassium ferricyanide chemiluminescence reaction by flow injectiion analysis. The pretreatment of samples in serum and the influence on concentration of reagents was investigated individually, there are good linear determination ranges, both column reactors showed good operational stability for run over 100 times, RSD was in 3.8~5.2%.
