甲基强的松龙上调受损脊髓细胞内Hsp27蛋白的表达 被引量:8

Up-regulated Expression of Hsp27 in Injured Spinal Cord Cells with Methylprednisolone Treatment in Early Period

摘要 目的研究甲基强的松龙(MP)上调热休克蛋白27(Hsp27)对受损中枢神经元自我保护及修复机制的激发和增强作用,为进一步促进受损中枢神经元存活和纤维再生创造有利的条件。方法采用改良Allen重物打击模型,动物随机分为2组:对照组(脊髓损伤后注入和实验组同等量生理盐水),实验组(脊髓损伤后5min使用大剂量甲基强的松龙)。分别在脊髓损伤24h后行脊髓病理形态学观察,用免疫组织化学染色的方法观察脊髓细胞中Hsp27的表达。用SPSS软件对数据进行统计学处理。结果①在热休克蛋白27量的表达上,两组差别有统计学意义(P<0.01)。②MP治疗可明显改善损伤脊髓的病理形态。结论MP可使具有保护作用的Hsp27表达上调,从形态学角度提示了MP可能激发并增强受损中枢神经的自我保护及修复机制。 Objective To study methylprednisolone's (MP) effect of up - regulation of Hsp27 on provocation and enhancement of self - protection and self - repair to injured central nerves, and if the injured nerves can get protection in an early period, and therefore MP benefit injured central nerves'survival and regeneration. Methods The rats were randomly divided into two groups: acute spinal cord injury groups, acute spinal cord injury rats with MP treatment groups. Improved Allen's weight drop model of acute spinal cord injury was prepared to study the pathological changes of spinal cord. 24 hours later, the spinal cords were taken out and cut, then the pathological changes of spinal cord cells with HE staining and the expression of Hsp27 in spinal cord cells with immunohistochemical staining were observed respectively. The data were analysed with SPSS soft ware. Results (1)The ex- pression of Hsp27 which is a protective to central nerves in the MP treatment groups was up - regulated. The quantity of Hsp27 in spinal cord cells in two groups has a difference in statistics ( P 〈 0. 01 ). (2)The pathological changes of spinal cord cells were significantly improved in MP treatment groups. Conclusion The that high - dose methylprednisolone treatment in an early period induces the up - regulation of Hsp27 and it might provocate and enhance the self - protection and self - repair of the injured central nerves in view of morphology.
