甲基强的松龙上调受损视网膜节细胞内Hsp27蛋白的表达 被引量:3

THE UP-REGULATED EXPRESSION OF HEAT-SHOCK-PROTEIN 27(Hsp27) IN INJURED RETINA GANGLION CELLS WITH METHYLPREDNISOLONE TREATMENT

摘要 目的研究甲基强的松龙(MP)上调热休克蛋白(Hsp27)蛋白对受损中枢神经元自我保护及修复机制的激发和增强作用,为进一步促进受损中枢神经元存活和纤维再生创造有利的条件。方法建立中枢神经损伤模型(视神经眼眶内切断术),经或未经MP治疗的术后动物分别存活4、7 14、21、28 d,取视网膜,切片,Nissl染色观察视网膜节细胞(RGCs)形态和存活数量的变化,用免疫组织化学染色的方法观察节细胞层中Hsp27的表达(检测A值)。用SAS软件对数据进行统计学处理。结果MP治疗组可见受损RGCs在71、4 d的存活数量增多,有统计学意义(P<0.01),在受损RGCs表达增多的Hsp27表达更加强烈(检测A值)。RGCs存活率与A值在各个时间点的变化趋势相似。结论MP可使具有保护作用的Hsp27表达上调,从形态学角度提示了MP可能激发并增强受损中枢神经的自我保护及修复机制。 Objective To study methylprednisolone's effect of up-regulation of Hsp27 on provocation and enhancement of self-protection and self-repair to injured central nerves, and if the injured nerves can get protection in early period, and therefore benefit injured central nerves'survival and regeneration. Methods Optic nerve axotomy was used in the experiment. The animals survived for 4, 7,14,21,28 days respectively after surgery with and without MP treatment. The retinas were taken out and cut, then the number and morphological changes of RGCs with Nissl staining and the expression of Hsp27 (optic density) in ganglion cell layers with immunohistochemical staining were observed respectively. The data were analysed with SAS soft ware. Results The quantity of the surviving retina ganglion cells increased in the group with high-dose intravenous methylprednisolone treatment at 7th and 14th days( P 〈0.01 ).The expression of Hsp27 that has a protective to central nerve in the injury retina ganglion cells was up-regulated. What ' s more, the ratios of survival cells and the change tendency of optic density ( A ) were resemble. Conclusion The present results indicate that high-dose methylprednisolone treatment induces the up-regulation of Hsp27 and it might provocate and enhance the self-protection and self-repair of the injured central nerve in view of morphology.
