摘要
目的:探讨脐带血干细胞体外诱导分化为心肌样细胞的动作电位、持续性钙电流特性,以对其进行电生理学鉴定。方法:用密度梯度离心和贴壁法筛选脐带血单个核细胞,培养于DMEM低糖培养基,经5-氮杂胞苷诱导为心肌样细胞,在诱导的不同阶段使用膜片钳技术检测其动作电位、持续性钙电流,并以大鼠心室肌作为对照。结果:脐带血干细胞体外诱导分化为心肌样细胞具有心肌特异性动作电位、持续性钙电流,其心肌特性伴随诱导后培养时间延长日趋典型。结论:脐带血干细胞体外诱导分化为心肌样细胞具有心肌细胞的电生理特性。
Objective To study active potential (AP) and last calcium ion current (ICa-L) of the myocardiomyocyte-like cell differentiated from human cord blood stem/progenitor cell and to make the physiological assessment to these cells. Method Cord blood mononuclear cells(CB-MNCs) were obtained by gradient centrifugation using Ficoll- Hyclone and adherent culture and cultured on 25 mm^2- culture flask, and then were induced and differentiated into myocardiomyocyte-like cell by 10 μmol/L 5-azacytidine. AP and ICa-L of these cells were measured by patch clamp technique at 2, 4 and 6 week after induction. The rat ventricular myocardium were regarded as the control. Result The myocardiomyocyte-hke cell differentiated from human CB-MNCs might produce the myocardial particular action potential and last calcium ion current in vitro, whose characteristic became more and more typical accompaning with culture time extension after induction. Conclusion The myocardiomyocyte-hke cell differentiated from human CBMNCs has the electrophysiological charactistics which are possessed particularly by the myocardial cell, so these cells may be regarded as the myocardial cell.
