人脐血干细胞移植治疗大鼠心肌梗死的实验研究 被引量:3

摘要 目的利用人脐带血干细胞移植治疗大鼠心肌梗死,探索治疗心肌梗死的新方法。方法将清洁级Wistar大鼠50只,随机分为3组:A组(心梗对照组)10只;B组(脐带血注射组)20只;C组(人脐带血干细胞移植组)20只。成功建立心肌梗死模型30min后,C组于缺血区心肌组织注射人脐带血干细胞50μl,B组注射脐带血50μl.4周后行心功能和病理学检测,并进行比较。结果C组大鼠的左室前壁厚度、左室舒张末期内径、左室收缩末期内径、左室短轴缩短率及左室射血分数较A组和B组有明显改善,各组间差异均有统计学意义(P<0.05);其毛细血管密度明显增加。结论人脐带血干细胞移植治疗可改善心肌梗死大鼠的心功能。 Objective To investigate a novel method to cure acute myocardial ischemia(AMI)by human umbilical cord blood stem cells(HUCBSC)transplantation. Methods A total of 50 Wistar rats were randomly divided as three groups. At 30 rain after establishment of AMI, HUCBSC was transplanted into the ischemic myocardium in HUCBSC transplantation group(n= 20), while the same volume of cord blood was transplanted into the isehemie myoeardium in cord blood injection group(n=20). Myocardial ischemia control group(n=10)only received left coro nary artery ligation. Four weeks later, the examinations of cardiac function and pathology were conducted and the resuits were compared between three groups. Results The thickness of anterior left ventricular wall(LVAW) ,left ventricular internal diameter at end - diastole(LVDd), left ventricular internal diameter at end systole(LVDs) ,left ventricular fractional shortening(FS) and left ventricular ejection fractions(EF) in HUCBSC group improved significantly than those in other two groups(P〈0. 05). In addition, capillary density in HUCBSC group increased evidently. Conclusion Transplantation of HUCBSC can improve myocardial function.
