中耳胆脂瘤表皮生长因子受体的检测

摘要 采用免疫组织化学ABC法对30耳中耳胆脂瘤、11耳健康外耳道皮肤进行表皮生长因子受体(EGFR)定位,用酶联免疫电转移印迹技术(EITB)检测8耳中耳胆脂瘤、4耳健康外耳道皮肤中EGFR。结果:130耳中,29耳上皮、24耳成纤维细胞、单个核细胞及血管内皮细胞显示EGFR阳性,免疫着色主要定位于细胞膜及细胞浆,且从基底细胞层到角质层逐渐减弱;外耳道健康皮肤仅散在基底细胞和棘细胞及6耳血管内皮细胞呈弱阳性。28耳胆脂瘤标本中有6耳在170KD处出现阳性蛋白带,所有外耳道健康皮肤未见任何免疫反应阳性产物。 Immunohistochemical localization and immunobiochemical detection of epidermal growth factor receptor (EGFR) in middle ear cholesteatoma (MEC) and normal external auditory canal (EAC) were adopted with ABC and enzyme-linked immunoelectrotransfer blot (EITB) technique The results were as follows: 1 Of the 30 ears of the MEC group positive immunoreactivity localized mainly in cell membrane and cytoplasm was revealed throughout the whole epithelial layers of 29 ears, and fibroblasts and mononuclear cells as well as vascular endothelial cells in 24 ears. Only discrete basal cells and spinous cells of all cases and vascular enthothelial cells of six in eleven ears of EAC group showed faint immunoreactivity 2 A clearly immunoreactive protein band corresponding to the site of 170KD standard protein was found in six of eight ears of MEC, while none of all four ears of EAC showed positive immunoreactive protein band The possible role played by EGFR in MEC was discussed
