摘要
研究野木瓜皂甙(Stauntoniae Saponin,SS)提取物对人类白血病细胞系HL60增殖的影响,为中草药抗肿瘤提供实验依据.以不同浓度SS作用于HL60细胞,然后用MTT比色法测定它对HL602细胞增殖的影响.当浓度为18mg/L时,SS能明显地促进HL60细胞的增殖(P<0.01);浓度大于90 mg/L时,SS对HL60细胞增殖产生抑制作用;当浓度为2253mg/L时,抑制率高达63.15%(P<0.01).浓度大于18mg/L后,细胞增殖的抑制率与SS浓度呈线性相关,r=0.8928(P<0.001);SS的LC50为1652 mg/L.野木瓜皂甙提取物对HL60细胞的增殖有明显的双向调节作用,即在较小范围的低浓度区域内有促进HL60细胞增殖的作用,浓度增大后则改变为抑制作用;浓度越大,抑制作用越强.
To investigate the effects of Stauntoniae Saponin (SS) on the proliferation of Human acute promyelocytic leukemia cell line HL60. Methods: After treated with SS with different concentrations, the proliferation of HL60 were tested by MTr assay. Results: As in 18mg/L, SS could promote the proliferation of HL60 ( P 〈 0. 01 ). With its increasing concentration ( 0.090mg/mL), SS reversed its effect on HL60 significantly. The ratio of inhibition on proliferation of HL60 was up to 63.15% when the concentration was in 2253mg/L ( P 〈 0. 01 ). The ratio of inhibition was linear relation to the concentration of SS when concentration of SS was greater than 18mg/L r = 0. 8929 (P 〈 0. 001 ). And its LC50 was 1652 mg/L. Conclusion: SS has bidirectional regulating effects on the proliferation of HL60. With the increasing concentration, the inhibiting effect of SS on HL60 would be greater.
