摘要 为了单独用纤维素酶制备酵母菌原生质体,测定了几种商品纤维素酶制剂,发现日本产纤维素酶onozukaR-10和y-c效果最好.纤维素酶制剂制取酵母菌原生质体的效果取决于其中β-1,3-葡聚糖酶的含量,与几丁质酶关系不密切,与纤维素酶本身酶活力无关.纤维素酶加1%的吐温80可提高制备原生质体的效果.酵母菌细胞经巯基乙醇或木瓜蛋白酶以及Na2CO3预处理后用纤维素酶较容易制备原生质体,制备的原生质体能恢复胞壁再生增殖. Yeast is one of the most important microbes in industry. In order to preparethe yeast protoplasts by cellulase alone,several products of cellulase were investigated. It was found that the cellulase “onozuka” R-10and cellulase y-c indicated the most efficient among them for preparation of yeast protoplasts. The release of yEast protoplasts depended on the β-1, 3-glucanases in the cellulase products,it was not fit close to the activity of cbitinase, neither could it relate to the cellulase itself. The effectiveness of the protoplast release was improved by adding 1% tween 80 to the cellulase solution. The yeast cells pretreated with mercaptoethanol or papin and Na2CO3 could easily release the protoplasts in the cellulase solution, and these protoplasts could regenerate the cell walls and division.
