Effect of sonodynamic chemistry therapy on C6 glioma cells in vitro

摘要 目的 研究声动力化学疗法(SDT)对体外C6胶质瘤细胞作用效果,试图探索一种治疗脑胶质瘤的新方法。方法 采用MTT法观察SDT后的各时段、加不同氧活性物质(ROS)后的抑瘤率;流式细胞学检测C6细胞的凋亡率及细胞周期百分比;透射电镜观察C6细胞亚细胞结构变化。结果 SDT后抑瘤率显著增高;超声组也有一定的抑瘤率;血啉甲醚(HMME)组抑瘤率与对照组无明显差别。与SDT组比较,加NaN,抑瘤率显著降低,加过氧化氢酶及SOD抑瘤率亦较SDT组低,加甘露醇抑瘤率无明显变化。SDT后凋亡率升高,G0/G1期、G2/M期百分比降低,S期百分比升高、凋亡增殖比(APR)升高。透射电镜观察对照组C6细胞无明显变化,SDT组细胞呈现凋亡或坏死状态。结论 SDT能显著杀伤体外c6胶质瘤细胞。 Objective To study the cytotoxic effect of sonodynamic therapy (SDT) on C6 glioma cells in vitro, explore a novel method for glioma treatment. Methods The inhibition rate of C6 glioma cells at different time after SDT, and SDT combined with different kinds of reactive oxygen species scavengers was investigated by MTT assay. The apoptotic rate and cell circle of C6 glioma cells was detected by flow cytometry (FCM). Ultrastructure changes of C6 glioma cells in four groups after SDT were observed with transmission electron microscope (TEM). Results The inhibition rate of C6 glioma cells after SDT significantly increased; Ultrasound group was also effective; the inhibition rate of hematoporphyrin monomethyl ether (HMME) group had no significant difference compared with control group. Compared with SDT group, the inhibition rate of Sodium azide group decreased significantly, and the Catalase (CAT) group and Superoxide dismutase (SOD) group decreased too, but the glycerin group had no significant decrease. In the group treated with SDT, the apoptotic rate increased and the percentage of G0/G1 ,G/M decreased, the percentage of S increased and the apoptosis rate increased. Observed by TEM, the changes of C6 glioma cells in control group were not significant, but the cells in SDT group displayed morphological features of apoptosis or necrosis. Conclusions SDT can kill C6 glioma cells significantly in vitro.
