摘要
目的初步建立脑胶质瘤标本库,探讨其应用价值。方法对脑胶质瘤患者进行手术前采静脉血,提取的基因组DNA保存在4℃冰箱内。在肿瘤切除术中,获取部分新鲜肿瘤组织标本和正常脑组织标本,保存在液氮中。收集进行组织病理学检查后剩余的肿瘤组织,制作成石蜡标本保存。在患者术后1年开始对其进行随访。采用生物芯片技术,对与胶质瘤一些相关的分子生物学指标进行初步研究。结果全血中提取的基因组DNA和冻存的肿瘤组织标本中提取的总RNA其 OD260/280比值都在1.7-2.1之间,能够满足胶质瘤分子生物学研究的需要。结论初步建立的脑胶质瘤标本库能够为肿瘤的分子生物学研究提供标本和相关资料来源。
Objective To establish glioma bank and analyze its scientific value.Methods The blood samples were taken from glioma patients at the operation rooms before anesthesia,and the genomic DNA was purified in laboratory,then stored at 4℃.The glioma specimens were collected during operations and frozen at once,then stored in liquid nitrogen,ff available,samples of the corresponding normal brain tissue and clinical data were also gathered.After the pathologists completed the diagnostic procedures and verified that the sample would satisfy the diagnostic needs,the residual tumor tissues were embedded with paraffin.All cases were followed up for one year after operation.The specimens were used for molecular biologic research by biochip technique.Results Genomic DNA extracted from blood samples and total RNA from the frozen glioma specimens,whose OD 260/280 were between 1.7-2.1,was suitable for molecular biologic research.Conclusions The glioma bank may supply materials for the.molecular biologic research on gliomas.
