中药小茴香对肝硬化腹水大鼠利尿作用机制实验研究 被引量:20

摘要 目的探讨中药小茴香对肝硬化腹水大鼠的利尿作用及其作用机制。方法将健康的SD雄性大鼠随机分为正常对照组(A组,8只)和肝硬化腹水造模组,造模组随机分为生理盐水组(B组)、双氢克尿塞组(C组)、中药小茴香组(D组),每组11只。用药4周后,收集24h的总尿量。取血进行肝功丙氨酸氨基转移酶(ALT)和醛固酮(ALD)放免检测。取肝、肾组织固定,进行HE染色和一氧化氮合酶(NOS)免疫组织化学切片。结果(1)造模后,大鼠腹腔内可见大量腹水,肝脏表面凸凹不平,有大小不等的结节。组织学检测可见大量假小叶形成,门管区炎细胞浸润。(2)双氢克尿塞(C组)和中药小茴香(D组)对肝硬化腹水大鼠总排尿量有明显的促进作用,与B组比较,差异有统计学意义(P<0.05)。(3)肝功能检测表明,B、C、D组大鼠血清ALT水平均高于A组,差异均有统计学意义(P<0.05);D组ALT水平低于B组和C组,差异均有统计学意义(P<0.05)。(4)B、C、D组ALD水平均高于A组(P均<0.05);D组ALD水平降低,与B组和C组比较,差异有统计学意义(P<0.05)。(5)肝组织HE染色显示,D组假小叶明显减少,汇管区炎症明显减轻。(6)NOS免疫组化显示,B组和C组有阳性结果,D组为阴性。结论(1)中药小茴香对肝硬化腹水大鼠有明显的利尿消腹水、改善肝纤维化的程度及肝功能的作用;(2)中药小茴香具有降低肝硬化腹水大鼠ALD、NOS水平的作用。 Objective: To investigate diuresis mechanism of Foeniculum Vulgare Mill on liver fibrosis ascites in rats. Methods: Healthy SD male rats were divided into the normal group (group A, 8 rats) and the model group , the model group was divided into randomly the normal saline group (group B), the hydrochlorothiazide group (group C) and the Foeniculum Vulgare Mill group (group D), 11 rats for each group. After 4 weeks, total volume of urine were collected for 24 h. The serum was obtained in order to test alanine aminotransferase (ALT) and aldosterone (ALD). Liver and kidney were collected for HE staining and NOS immunohistochemistry slice. Results: (1) A lot of ascites were seen in abdominal cavity, inequality of size of tubercles could be seen on liver surface, a lot of pseudlobes and inflammation were observed by histological examination after induced. (2) The total volume of urine in group C and group D was more than group B. Compared with group B, both C and D groups had significant difference (P〈0. 05). (3) The ALT in group B,C and D were higher than that of group A, the difference was significant. The ALT in group D was lower than that of group B and C and the differences were also significant (P 〈0.05). (4) The ALD in group B,C and D were higher than that of group A, the differences were significant. The ALD in group D was lower than that of group B and C and the difference was significant (P 〈0.05). (5) HE staining of liver indicated that pseudlobes and inflammation were decreased obviously in group D; (6)The results of NOS indicated that both group B and C were positive, group D was negative. Conclusion: (1) Chinese herb Foeniculum Vulgare Mill could accelerate diuresis, improve the degree of fibrous liver and reverse the function of liver. (2) Chinese herb Foeniculum Vulgare Mill could decrease the levels of ALD and NOS of liver fibrosis ascites rats.
