摘要 目的观察超抗原SEA体外对小鼠脾淋巴细胞活化和增殖的影响。方法SEA体外诱导小鼠脾淋巴细胞,测定淋巴细胞、T细胞、B细胞的增殖能力,用流式细胞术检测淋巴细胞膜表型的变化,并测定脾淋巴细胞培养上清中IL-2、IL-4、IL-10、INF-γ的水平。结果不同浓度SEA诱导脾淋巴细胞的增殖能力不同,在1~4d内的增殖变化也不完全一样,SEA浓度为103ng/mL时,脾淋巴细胞的活化和增殖能力最强。SEA对T细胞(无APC)和B细胞的增殖无明显影响。细胞培养上清中IL-2和INF-γ的水平显著升高,IL-4和IL-10含量低。结论在APC递呈作用下,SEA能显著诱导初始CD4+T细胞和CD8+T细胞的活化和增殖,促使TH0细胞向TH1细胞分化,分泌IL-2、IFN-γ等细胞因子。 Objective To observe the effects of SEA on the activation and proliferation of spleen lymphocytes in vitro. Methods Examined the proliferation and surface molecule expression of the spleen lymphocytes induced by SEA in vitro. Observed the difference of effects on spleen lymphocytes,T cells or B cells induced by SEA . And measured the production of IL-2,IL-4,IL- 10,INF-γ in the supernatant. Results There were different effects on the proliferation of spleen lymphocytes induced by different doses of SEA, and 10^3 μg./L was the most efficient dose of SEA . There were no significant effects on the proliferation of T cells ( without APC) and B cells. The production of IL - 2, INF - γ in the supernatant were increased significantly, but the production of IL -4,IL - 10 were very low. Conclusion The activation and proliferation of CD 4^+ and CD8^+ naive T cells presented by APC could be induced strongly by SEA. Which helped Th0 cells differentiate to Thl cells and enhanced IL- 2 and INF-γ production.
