华蟾素诱导人红白血病细胞K562凋亡及对Caspase-3表达的影响 被引量:18

摘要 目的观察华蟾素对白血病细胞的凋亡诱导作用并探讨其机制。方法通过MTT法观察华蟾素对K562细胞的生长抑制作用。通过DNA电泳、流式细胞术、AnnexinV标记法检测华蟾素作用后K562细胞的凋亡状况。流式细胞术分析Caspase-3表达的影响。结果华蟾素能明显抑制K562细胞生长,且有浓度依赖关系,作用48小时的IC50值为(0.152±0.012)ug/ml。K562细胞经0.5ug/ml的华蟾素作用48小时后,DNA琼脂糖电泳可见凋亡的特征性梯形条带。0、0.0625、0.125、0.25、0.5ug/ml的华蟾素作用K562细胞48小时后的凋亡率分别为(3.78±0.09)%、(3.84±0.19)%、(39.43±1.90)%、(64.55±2.70)%、(81.05±4.23)%。华蟾素作用后K562细胞Caspase-3表达增高在0.0625±0.5ug/ml的浓度范围内具有一定的量效关系。结论华蟾素对红白血病细胞K562具有凋亡诱导作用,其机制与提高Caspase-3表达有关。
