反相高效液相色谱法同时测定乳酸及乳酸甲酯 被引量:20

Simultaneous Determination of Lactic Acid and Methyl Lactate by Reversed-Phase High Performance Liquid Chromatography

摘要 乳酸及乳酸甲酯是重要的有机化工原料。在由乳酸合成乳酸甲酯的绿色化工工艺研究过程中,为了优化反应条件,建立了一种同时分离测定乳酸连续酯化反应体系中乳酸和乳酸甲酯的反相高效液相色谱法。HPLC分析条件为:Prevail C18色谱柱,以V[NaH2PO4/H3PO4缓冲液(pH2.0)]:V(甲醇)=90:10为流动相,流速为1.0mL·min^-1,检测器为示差折光检测器。在进样量为0.2~20μg成良好的线性关系。乳酸的加样回收率为94.55%~106.94%,相对标准偏差为1.23%~2.01%;乳酸甲酯的加样回收率为91.03%~109.0%,相对标准偏差为1.25%~2.88%。该方法准确、简捷、可靠。 During the research ot the green chenncal techmque tor conversion ot lactic acid to methyl lactate, a new reversed-phase high performance liquid chromatographic (RP- HPLC ) method was developed for the simultaneous determination of lactic acid and methyl lactate in the continuous esterification system in order to optimize the reaction conditions. The HPLC conditions were Prevail C18 column with a mixture of V[ NaH2PO4/H3PO4 buffer ( pH 2. 0): V( methanol ) = 90:10 as the mobile pase at a flow rate of 1.0 mL/min and with refractive index detector. The linear range of lactic acid and methyl lactate were both 0. 2 -20 μg. Recovery of lactic acid was 94. 55% - 106. 94% with relative standard deviations 1.23% - 2. 01% ( n = 3 ). Recovery of methyl lactate was 91.03% - 109. 0% with relative standard deviations 1.25% - 2. 88% (n = 3). This method is simple,rapid and accurate.
