摘要
目的:对自制的、用于癌性腹水的中药复方制剂———消水Ⅱ号中的两种药材冰片和桂枝中有效成分进行透皮吸收的定量或定性分析,初步探讨该药的在体透皮吸收行为。方法:以龙脑/异龙脑和桂皮醛作为消水Ⅱ号有效成分透皮吸收的指标成分。在H22荷瘤小鼠腹部皮肤上敷用消水Ⅱ号药膏,用气相色谱法和高效液相色谱法分别测定给药后不同时间内小鼠腹水中龙脑/异龙脑和桂皮醛的含量。结果:气相色谱测定图谱显示,在敷药后15分钟时,在小鼠腹水中尚不能测出龙脑/异龙脑;但在药后30分钟,腹水中龙脑/异龙脑的含量已达到1.707μg/mL;达峰值(9.588μg/mL)时间约为4 h。HPLC图谱显示,敷药2小时后,能在腹水中测到有桂皮醛的存在。结论:结果表明,合理配制的中药复方制剂可以经皮肤给药,其有效成分能透皮吸收进入体内。同时,结果也初步认定,气相色谱法和高效液相色谱法可用于消水Ⅱ号中有效成分透皮吸收量的检测,且有准确、简便、快速等优点。
Objective： Ⅱ Recipe of Eliminating Effusion Ointment which was formulated by us is a kind of Traditional Chinese Medicine for eliminating tumor effusion. To probe the absorbed character of Ⅱ Recipe of Eliminating Effusion Ointment, two effective ingredients in this formula by percutaneous absorption were determined. Methods： In thistest, the concentrations of bomeol/isobomeol and cinnamyl-adehyde in model mouse ascites were as signs of this formula permeating skin after external application. Heptoma ascites line （ H22 ） mice were given Ⅱ Recipe of Eliminating Effusion Ointment for external application on abdominal skin, and then, at different time, the concentrations of cinnamyl-adehyde and bomeol/isobomeol in mouse ascites were separately detenrtined by gas chromatography （GC）or HPLC. Results： The chromatograms of GC showed that bomeol/isobomeol in ascites can not be determined within 15 min, after being given the medicine. But at 30 min, the concentration has reached 1.707μg/mL. The peak value （9.588μg/mL） is at 4 h after being given Ⅱ Recipe of Eliminating Effusion Ointment. The chmmatograms of HPLC showed that cinnamyl-adehyde in model mouse ascites is got at 2 h after application. Conclusion： The results indicate that the main effective ingredients of Ⅱ Recipe of Eliminating Effusion Ointment can be absorbed through mice＇ skin. And the determinative methods used in this test were efficient and accurate. The methods can be used for the further percutaneous absorption research on the Ⅱ Recipe of Eliminating Effusion Ointment.
