摘要
目的:探讨补中益气汤不同浓度、不同时间对脾气虚证大鼠胃泌素(gastrin,GAS)基因表达的影响。方法:以大黄法、游泳力竭法、饥饿法三因素复合的方法复制脾气虚证大鼠模型,造模大鼠随机分成模型组、补中益气汤低、中、高剂量组,在干预后不同时间(7d,14d,21d),用逆转录聚合酶链反应方法(RT-PCR)检测补中益气汤对胃窦GAS mRNA表达的影响。结果:与模型组比较,补中益气汤中、高剂量组血清GAS水平升高(P<0.05,P<0.01)、胃窦GAS mRNA表达上调(P<0.05,P<0.01);中、高剂量组比较,差异有显著性意义(P<0.05),以中剂量组为佳。结论:补中益气汤能上调GAS mRNA表达可能是其治疗脾气虚证的机制之一。
Objective: To explore the effect of bushong yiqi decoction (BZYQD) with various concentrations at different time points on gastrin (GAS) gene expression level in spleen-qi deficiency rats. Methods: The splean-qi deficiency rats were randomly divided into the model control group and the low, medium and high dosage of BZYQD groups. Reverse transcriptase polymerase chain reaction was used to observe the effect of BZYQD on the GAS mRNA of gastric antrum in rats treated at different time points (7, 14, 21d). Results: As compared with that in model control group, the GAS gene expression level were significantly higher both in the medium and high dosage groups ( P 〈0.05, P 〈0.01). And the increment in the medium dosage group was significantly higher than that in the high dosage group ( P 〈 0.05). Conclusion: Up-regulation on GAS gene expression is possibly one of the mechanisms of BZYQD in treating spleen-qi deficiency rats.
