摘要
目的 探讨褪黑素(melatonin,Mel)对匹罗卡品(pilocarpine,PILO)致痫大鼠行为及脑电图改变的影响。方法 通过腹腔注射(intraperitoneal injection,i.p)匹罗卡品建立癫痫动物模型。25只雄性Wistar大鼠分为5组,即对照组、PILO组、Mel10mg/kg组、Mel25mg/kg组、Mel50mg/kg组,每组5只大鼠。观察大鼠行为、脑电图及海马组织学改变。结果腹腔注射Mel10,25,50mg/kg均可延长痫样放电的潜伏期、降低痫波发放频率、减轻痫性发作及海马损伤程度。尤其是Mel25mg/kg组与PILO组比较痫性放电潜伏期[(23.6±5.32)min VS(16,4±2.70)min]明显延长(P〈0.05),痫波发放频率[(92.8±8.11)次/min VS(122.6±13.28)次/min]显著降低(P〈0.01)。结论 Mel在PILO致痫大鼠模型中具有抗惊厥作用,该作用具有一定的剂量依赖性。
Objective To probe the effect of melatonin on behavior and electroencephalograph(EEG) in epileptic rats induced by pilocarpine (PILO). Methods Use model of epilepsy induced by pilocarpine; twentyfive male Wistar rats were divided into five groups, control group, PILO group, melatonin 10 mg/kg intraperltoneal injection(i, p. )group, melatonin 25 mg/kg i.p. group, melatonin 50 mg/kg i.p. group, each group had five rats. To observe changes of EEG recording, behavior and hippocampal histology. Results The latency to the appearance of first epileptic discharge was significantly increased, while the frequency of epileptic discharge, serious degree of seizure and hippocampus damage were significantly decreased when the epileptic rats were administered with melatonin 10 - 50 mg/kg, especially in Melatonin 25mg/kg i.p. group compared to PILO group. Conclusion melatonin has the role of anti-convulsion in epileptic rats induced by PILO, the effect is dose dependent, displaying maximal activity when melatonin 25 mg/kg was injected.
